My favorite deer caliber is the .25-06 Remington. Other good deer calibers are: .270 Winchester, .308 Winchester and .30-06 Springfield.
The minimum deer caliber is the .243 Winchester and up to say about a 7mm Remington Magnum without getting into 'over-kill.' For bigger game say in North America you don't want anything smaller than the .308. If you're going to hunt on the Continent (Africa) than the .375 Holland and Hollang Magnum is considered the minimum for dangerous game.

For medium game like deer, 7x57 Mauser is hard to beat, even 106 years after its introduction. If you just have to have something more modern, 7mm08 is nearly as good. For all big game, the single rifle would have to be 375 H%26amp;H or the new 375 Ruger. And in a three-gun battery, either that or 9.3x62 (or 9.3x64) would be a good middle gun, with 450 Rigby or something similar on the large end, retaining the 7mm as the small rifle. Add a 22 and a shotgun, and you'd be well equipped for any game, anywhere.
What type of deer?
I use a .300WM on fallow and its sometimes a bit messy, I only have this calibre so I can also hunt Reds and Sambar etc without having to own 2 rifles. I have a .243 but its always stressing me, if I don't place the shot spot on I will have a runner on my hands.
If I could own 2 rifles it would be a .270 for the Fallow and the .300 for Sambar deer. I like the faster flatter rounds, just my preference.
Its an age old argument my friend and you just had to go and ask it LOL.
I have friends who swear by their 30-06 and others who wouldn't use anything other than .270 (on fallow).
In the end its your choice but remember you want em dead, not doing a runner because you used something too small.
When you say big game you immediately start talking .338 Win or up to and above .375 H%26amp;H

Do you mean 2 rifles? One for deer and another for big game? If its 2 rifles then get a rifle chambered in .270 for deer and 30/06 or 300 Winchester mag for big game... I wouldnt go too much bigger -- In my opinion you start getting to the point of diminishing return once you start moving into bigger calibers than .300 win mag...... If your a regular working kinda of guy with a regular budget then the Remington 700 bolt actions are nice. Also Savage makes excellent rifles....... If you have more $$$ than the average joe then Kimber is the way to go..... See below....
all around, 30-06 or 308 would be hard to beat. these are two of the most popular calibers out there. they will handle just about everything in north america without a fuss. unless you plan to hunt kodiak brown or polar bear, then i would find something a little bigger, like the 300 mag.
personally, i like the 7mm-08 or 7mm mauser for deer, and these will handle most other big game as well, bears excepted.
if I could only have one rifle it would be a 300 win mag.
if I could have two rifles one would be a 280 remington shooting 140 grain accutips for deer or similar size critters up to black bear or even elk. With the 300 mag with nosler partition bullets for moose , and large bear. In fact I do own both of these and I use my 280 more than any other rifle I own.

The 30-06 is the best choice.* You have a greater variety of bullet weights to choose from.* It will harvest any game in North America cleanly and efficiently.* I prefer the 180 or 220 grain myself.*
For deer, it's hard to beat the venerable .30-06, or a .30-30 for shorter shots. For bigger game, look into a 7MM Mag or 300 Magnum.
Corbin, the .270 Win is your best bet. I own several in Remington 700 actions and they are wonderful. You can ';accurize'; and improve upon your rifle as you wish. Enjoy!
id say the 270. I have one on a remington 700 mountain rifle. Its light and will do the job nicely.
